Anticancer effects of garlic and garlic-derived compounds for breast cancer control.

Garlic and garlic-derived compounds reduce the development of mammary cancer in animals and suppress the growth of human breast cancer cells in culture. Oil-soluble compounds derived from garlic, such as diallyl disulfide (DADS), are more effective than water-soluble compounds in suppressing breast cancer. Mechanisms of action include the activation of metabolizing enzymes that detoxify carcinogens, the suppression of DNA adduct formation, the inhibition of the production of reactive oxygen species, the regulation of cell-cycle arrest and the induction of apoptosis. Selenium-enriched garlic or organoselenium compounds provide more potent protection against mammary carcinogenesis in rats and greater inhibition of breast cancer cells in culture than natural garlic or the respective organosulfur analogues. DADS synergizes the effect of eicosapentaenoic acid, a breast cancer suppressor, and antagonizes the effect of linoleic acid, a breast cancer enhancer. Moreover, garlic extract reduces the side effects caused by anti-cancer agents. Modulation of histone acetylation by garlic sulfur compounds.

Preclinical studies have shown that fresh garlic extracts, aged garlic, garlic oil and specific organosulfur compounds generated by processing garlic could alter carcinogen metabolism, inhibit tumor cell growth through induction of cell cycle arrest or apoptosis, or angiogenesis. In particular, recent studies have suggested that anticarcinogenic effects of certain garlic compounds may implicate at least in part a modulation of histone acetylation, a process involved in the regulation of gene expression, resulting from the inhibition of histone deacetylase activity. The aim of this review is to describe available data on sulfur compounds from garlic and histone acetylation and to discuss their potential for cancer prevention. Available data indicate that garlic compounds could inhibit histone deacetylase activity and induce histone hyperacetylation in vitro as well as in vivo. Sparse studies provide evidence of an involvement of histone acetylation in modulation of gene expression by diallyl disulfide and allyl mercaptan. These effects were observed at high concentrations. Organic sulfur compounds (OSCs) derived from plants, fungi or bacteria can serve as chemopreventive and/or chemotherapeutic agents and have been attracting medical and research interest as a promising source for novel anti-cancer agents. Garlic, which has long been used as a medicinal plant in different cultures due to its multiple beneficial effects, contains a consistent number of OSCs, the majority of which are currently under investigation for their biological activities. Experimental animal and laboratory studies have shown strong evidence that garlic OSCs may affect cancer cells by promoting early mitotic arrest followed by apoptotic cell death without affecting healthy cells. The ability of OSCs to hinder cancer cell proliferation and viability tightly correlates with the length of the sulfur chain. Current data support a mechanism of mitotic arrest of cancer cells due to the alteration of the microtubule network, possibly as a consequence of the high reactivity of sulfur atoms against the thiol groups of different cellular macromolecules controlling crucial regulatory functions. The Ministry of Defence turned large parts of the country into a giant laboratory to conduct a series of secret germ warfare tests on the public. A government report just released provides for the first time a comprehensive official history of Britain’s biological weapons trials between 1940 and 1979. Many of these tests involved releasing potentially dangerous chemicals and micro-organisms over vast swaths of the population without the public being told. While details of some secret trials have emerged in recent years, the 60-page report reveals new information about more than 100 covert experiments. The report reveals that military personnel were briefed to tell any ‘inquisitive inquirer’ the trials were part of research projects into weather and air pollution. The tests [were] carried out by government scientists at Porton Down. In most cases, the trials did not use biological weapons but alternatives which scientists believed would mimic germ warfare and which the MoD claimed were harmless. But families in certain areas of the country who have children with birth defects are demanding a public inquiry.–

Note: Military personnel were ordered to lie to cover-up potentially dangerous experiments on the public. So how can we trust that these people have the public interest as a priority?

Green tea extract selectively targets nanomechanics of live metastatic cancer cells.

Green tea extract (GTE) is known to be a potential anticancer agent (Yang et al 2009 Nat. Rev. Cancer 9 429-39) with various biological activities (Lu et al 2005 Clin. Cancer Res. 11 1675-83; Yang et al 1998 Carcinogenesis 19 611-6) yet the precise mechanism of action is still unclear. The biomechanical response of GTE treated cells taken directly from patient’s body samples was measured using atomic force microscopy (AFM) (Binnig et al 1986 Phys. Rev. Lett. 56 930). We found significant increase in stiffness of GTE treated metastatic tumor cells, with a resulting value similar to untreated normal mesothelial cells, whereas mesothelial cell stiffness after GTE treatment is unchanged. Immunofluorescence analysis showed an increase in cytoskeletal-F-actin in GTE treated tumor cells, suggesting GTE treated tumor cells display mechanical, structural and morphological features similar to normal cells, which appears to be mediated by annexin-I expression, as determined by siRNA analysis of an in vitro cell line model. Our data indicates that GTE selectively targets human metastatic cancer cells but not normal mesothelial cells, a finding that is significantly advantageous compared to conventional chemotherapy agents.–PMID: 21451222 [PubMed – indexed for MEDLINE]

 

Recipe For making a Green Tea Extract—take green tea leaves ( get the gunpowder if you can from china it is rolled up during harvest making it ka more potent green tea as well ) take3-4 tablespoons of the leaves and add to a blender then add 1 -1/2 cup of alcohol clear or brandy or wine—then put lid on and blend for 10 minutes and stop blender and strain through a handkerchief –this is one method—be aware that the solution maybe hot so even if you wait til it is cool or til you can handle –safely strain out the green tea through the handkerchief—this is an extract—to make it more potent take the extract and pour a portion into a pot and simmer it down to about half to ¼-1/2 of where you started–Then allow to cool and the bottle—this solution should be good at 3-6 drops 2 times a day—now this is an ultra potent extract

‘It has to go away’: Facebook director calls for an end to internet anonymity

 

Randi Zuckerberg, marketing director of Facebook has spoken out against internet anonymity in a bid to end cyber bullying Mrs Zuckerberg argued the end of on-line anonymity could help curb the trend of trolling and harassment on the web.–Speaking at a Marie Claire panel discussion on social media, she said: ‘ I think anonymity on the Internet has to go away.[U1]–‘People behave a lot better when they have their real names down. … I think people hide behind anonymity and they feel like they can say whatever they want behind closed doors.'[U2] The comments echo those of former Google CEO Eric Schmidt who has previously labelled internet anonymity a ‘dangerous’ precedent, before predicting government intervention will one day lead to its demise. Controversial: Mark Zuckerberg has recently hit the headlines for proposing to allow under 13s on Facebook –Concerned: Former Google CEO Eric Schmidt has also said previously internet anonymity would possibly come to an end Privacy advocates have however condemned previous attempts to dismantle on-line anonymity. –attacked: Erin Andrews was also present at the meeting to speak about her traumatic experience of cyber bullying—[U3]Critics complain that the forced introduction of some kind of ‘on-line passport’ would damage the freedom of speech and blunt the internet as a tool for dissidents to speak up against oppressive governments.–The panel also pressed Mrs Zuckerberg to list what forthcoming safeguards the site has to protect security, she declined.–She added simply: ‘There’s so much more we can do.—‘We’re actively tying to work with partners like Common Sense Media and our safety advisory committee.’–

The panel were joined by Erin Andrews, an ESPN anchor who had a naked video of her posted on-line by a stalker[U4].–At times becoming emotional, Mrs Andrews asked Mr Schmidt why searches for her name still brought up images and videos of the offending tape, despite her repeated attempts to have them removed.

Adding that just because she was a woman in her 30s She said: ‘It’s still cyber bullying.–‘Somebody needs to step in. As a family we’re always asking, what is it going to take.’–Facebook has previously come under fire after founder Mark Zuckerberg said he would ‘fight’ to allow under-13s to use the social networking website – despite warnings it would put the most vulnerable children at risk.–

In America the age limit, also of 13, is dictated by laws designed to protect young children, but Facebook’s 27-year-old billionaire creator believes the educational benefits of using the site mean the restrictions should be lifted.[U5]

A damning report co-authored by an international group of scientists and

researchers for non-government organisation Open Earth Source (OES) reveals that studies from industry including those from Monsanto since the 1990s showed glyphosate caused birth defects and the European Commission approved the herbicide in full knowledge of those finding [1]. —Glyphosate is the active ingredient in Monsanto’s Roundup herbicide and over a hundred other commercial formulations. It is the cause of great concern as evidence of its harmful effects keep piling up from independent scientific studies in recent years, including endocrine disruption, DNA damage, reproductive and developmental toxicities, neurotoxicity, cancer, and birth defects (see [2] Glyphosate Toxic and Roundup Worse, SiS 26; [3] Death by Multiple Poisoning, Glyphosate and Roundup, SiS 42; [4] Ban Glyphosate Herbicide Now. SiS 43; [5] Lab Study Establishes Glyphosate Link to Birth Defects, SiS 48).———–

Today, almost 90 percent of the world’s soy production takes place in the US, Brazil and Argentina, and in 2009, RR soybean accounted for 91 percent, 99 percent and 71 percent respectively of total soybean acreage in those countries. ———Since 1997, RR soy production has increased from 5 to 30 million hectares of land in the US alone [1]. Soybeans have been found to contain glyphosate residues up to 10 times as high as the doses that caused foetal malformations in chick and frog embryos.——-With such widespread use of the herbicide, and the EU considering approval of GM crops tolerant to glyphosate for cultivation in Europe, there is an urgent need for a proper review of the herbicide, which is in line with the more stringent new EU pesticide regulation that came into force in June 2011. Indeed, just such a review was due to take place in 2012. But shortly after the EC was notified of the latest research on glyphosate causing birth defects (which the EC dismissed), it delayed the review on glyphosate and 38 other dangerous pesticides to 2015 [U6][1]. Furthermore, the 2010 review will be under the old, less stringent regulations. The official reason for the delay is that they have ‘too much workload’. This means that the safety of glyphosate may not be reviewed under the new regulations until 2030. —The review delay is being challenged in a lawsuit brought against the EC by Pesticides Action Network Europe and Greenpeace. The co-authors of the OES report are also calling on the EC to conduct a prompt review without delay, and to withdraw glyphosate and Roundup from the market.

 

Evidence of birth defects from industry and independent studies The OES report

reveals that studies carried out by industry in the 1990s showed embryonic lethality and birth defects in laboratory animals, including dilation of the heart in rabbits at low doses of glyphosate. Higher doses were shown to cause defects in independent studies as far back as 1980. —Monsanto has responded by denying its own findings [6]: “Regulatory authorities and independent experts around the world agree that glyphosate does not cause adverse reproductive effects in adult animals or birth defects in offspring of these adults exposed to glyphosate, even at doses far higher than relevant environmental or occupational exposures.”[U7]However, the ‘independent experts’ cited by Monsanto were mired in conflicts of interests, both financial and professional, and rely almost entirely on studies carried out by industry [1]. —Independent studies have revealed links to cancer, genetic damage and endocrine disruption, as well as developmental defects, for example, craniofacial and vertebrae abnormalities in rats [7] as well as craniofacial and mouth deformities, eye abnormalities and bent, curved tails in frog tadpoles [8]. The most recent study by Carrasco and colleagues found a mechanistic link between glyphosate and abnormal retinoic acid signalling in embryonic development [5, 9] ——-Astonishingly, these and numerous other studies cited by the OES report were dismissed by the European Commission (EC) and the German Federal Office for Consumer Protection and Food Safety (BVL), which is responsible for liaising between industry and the EC. BVL’s conclusion, communicated to the EC via Germany’s 1998 ‘draft assessment report’ (DAR), was ‘‘no evidence of teratogenicity” for glyphosate.

The neural networks in the brains of the middle-aged and elderly have weaker connections and fire less robustly than in youthful ones–ScienceDaily (July 28, 2011) — Yale University researchers can’t tell you where you left your car keys — but they can tell you why you can’t find them.—A new study published July 27 in the journal Nature shows the neural networks in the brains of the middle-aged and elderly have weaker connections and fire less robustly than in youthful ones. Intriguingly, the research suggests that this condition is reversible.—“Age-related cognitive deficits can have a serious impact on our lives in the Information Age as people often need higher cognitive functions to meet even basic needs, such as paying bills or accessing medical care,” said Amy Arnsten, Professor of Neurobiology and Psychology and a member of the Kavli Institute for Neuroscience. “These abilities are critical for maintaining demanding careers and being able to live independently as we grow older.”—As people age, they tend to forget things more often, are more easily distracted and disrupted by interference, and have greater difficulty with executive functions. While these age-related deficits have been known for many years, the cellular basis for these common cognitive difficulties has not been understood. The new study examined for the first time age-related changes in the activity of neurons in the prefrontal cortex (PFC), the area of the brain that is responsible for higher cognitive and executive functions.—Networks of neurons in the prefrontal cortex generate persistent firing to keep information “in mind” even in the absence of cues from the environment. This process is called “working memory,” and it allows us to recall information, such as where the car keys were left, even when that information must be constantly updated. This ability is the basis for abstract thought and reasoning, and is often called the “Mental Sketch Pad.” It is also essential for executive functions, such as multi-tasking, organizing, and inhibiting inappropriate thoughts and actions. Arnsten and her team studied the firing of prefrontal cortical neurons in young, middle-aged and aged animals as they performed a working memory task. Neurons in the prefrontal cortex of the young animals were able to maintain firing at a high rate during working memory, while neurons in older animals showed slower firing rates. However, when the researchers adjusted the neurochemical environment around the neurons to be more similar to that of a younger subject, the neuronal firing rates were restored to more youthful levels.—-Arnsten said that the aging prefrontal cortex appears to accumulate excessive levels of a signaling molecule called cAMP, which can open ion channels and weaken prefrontal neuronal firing. Agents that either inhibited cAMP or blocked cAMP-sensitive ion channels were able to restore more youthful firing patterns in the aged neurons. One of the compounds that enhanced neuronal firing was guanfacine, a medication that is already approved for treating hypertension in adults, and prefrontal deficits in children, suggesting that it may be helpful in the elderly as well.—Arnsten’s finding is already moving to the clinical setting. Lyme Disease Tick Adapts to Life On the (Fragmented) Prairie

ScienceDaily (June 22, 2011) — A new study offers a detailed look at the status of Lyme disease in Central Illinois and suggests that deer ticks and the Lyme disease bacteria they host are more adaptable to new habitats than previously appreciated.–Led by researchers at the University of Illinois, the study gives an up-close view of one region affected by the steady march of deer ticks across the upper Midwest. Their advance began in Wisconsin and Minnesota and is moving at a pace of up to two counties a year in Illinois and Indiana.—Today the deer tick is established in 26 Illinois counties, up from just eight in 1998, said Illinois Department of Public Health entomologist Linn Haramis. Reports of human Lyme disease cases in the state have more than tripled in the same period[U1], he said.–“We’ve had several years in a row where we’ve had over 100 cases, up from about 30 per year more than 10 years ago,” Haramis said. “It’s not a huge increase, but it’s been steady and there’s an upward trend.”—Deer ticks are known to do best in forested areas, where they can readily move from small mammals (which provide their first meal) to moist leaf litter on the forest floor, and then to deer, on which they mate. Deer ticks do not pick up the Lyme infection from deer, said Jennifer Rydzewski, who completed her master’s degree with the study in the department of natural resources and environmental sciences at the University of Illinois.–“The deer tick will feed on a variety of mammals, birds and even reptiles,” she said. “But Borrelia burgdorferi, the bacterium that causes Lyme disease, replicates really well within white-footed mice, so white-footed mice are the main reservoir that passes that bacterium on to the immature ticks that are feeding on it.”–White-footed mice also are forest dwellers. Prior to the new study, little was known about whether, or how, Lyme disease persists in other habitat types.—To determine if Lyme disease had gained a foothold in the patchwork of forests, farms and prairies of Central Illinois, researchers trapped small mammals in Allerton Park, a 1,500-acre (600-hectare) natural area in Piatt County. They focused on four habitat types: young forest, mature forest, a flood plain and a 30-acre (12-hectare) patch of prairie surrounded by woods and agricultural fields.—The researchers removed deer ticks from the mammals they trapped and tested the ticks for Lyme disease. They found that the immature forest and the prairie hosted the highest percentage of deer-tick-infested mammals, the highest number of ticks per mammal trapped and the highest rates of ticks infected with Lyme disease of the four habitat types evaluated.—“The highest prevalence of B. burgdorferi infection was found (in deer tick larvae) from the prairie (27 percent) followed by the young forest (15 percent), the mature forest (6 percent) and the flood plain (6 percent),” the researchers wrote.–[U2]”Interestingly, all of the positive ticks from the prairie were from prairie voles, not the typical white-footed mouse,” Rydzewski said. There also were many more ticks per animal on the prairie voles than on the white-footed mice of the forest, she said.

This is the first study to report evidence that the prairie vole may potentially serve as a competent reservoir host for the Lyme disease bacterium, B. burgdorferi, said Nohra Mateus-Pinilla, a wildlife veterinary epidemiologist at the Illinois Natural History Survey who led the study with Rydzewski and natural resources and environmental sciences emeritus professor Richard Warner. (The Survey is a unit of the Prairie Research Institute at Illinois.)–“The fact that we found tick larvae feeding so prominently on prairie voles and those ticks were infected and hadn’t had a chance to feed on anything else is a very strong indicator that we are dealing with a different reservoir of Lyme disease that deserves more attention,”[U3] Mateus-Pinilla said.—The researchers hypothesize that when newly hatched ticks find themselves on the prairie, they latch on to the first small mammal that comes along, which in most cases is a prairie vole (white-footed mice prefer the forest). The abundance of prairie voles in the prairie is much lower than that of the white-footed mice in the forest, so more tick larvae and nymphs end up on the same few prairie voles. Since the number of ticks per animal is higher on the prairie, the likelihood of infection is higher there as well.—“The landscape of Illinois, especially the northern and central area, is very fragmented with agricultural and other development, so there aren’t really big continuous areas that are forested,” Rydzewski said. “And so maybe these ticks are finding new habitats to establish themselves in because of the lack of previous habitats.””What’s exciting about the new findings is that we are dealing with potentially new mechanisms of disease transmission that we just have not explored and perhaps we do not understand,”[U4] Mateus-Pinilla said. “We need to think outside of what we already know about Lyme disease transmission.”–The new study appears in the journal Vector-Borne and Zoonotic Diseases. White blood cells from mice that are naturally immune to cancer cured tumors in other mice and provided them with lifelong immunity to the disease, researchers reported Monday. The finding indicates the existence of a biological pathway previously unsuspected in any species. A small team of researchers is working to understand the genetic and immunological basis of the surprising phenomenon. Preliminary studies hint at the existence of a similar resistance in humans. Researchers hope that harnessing the biological process could lead to a new approach to treating cancer. But Dr. Zhen Cui of Wake Forest, whose team published the findings in the Proceedings of the National Academy of Science, said he expected rapid replication of the results because the findings were so clear-cut and easily observed. “This is a truly remarkable phenomenon — and it really needs confirmation from other institutions,” he said. The team took white blood cells from the immune mice … and injected them into mice already carrying a variety of tumors, some of which were extremely aggressive. In every case, the cancers were destroyed, even if the cells were injected at a point distant from the tumor. Healthy tissues were not affected. The mice that received the cells, furthermore, were protected from new tumors for the rest of their lives. The researchers have no idea how the immunity continues.
